Overview

[44Sc]Sc-B11 nanobody is a research-stage radiolabeled imaging probe designed for positron emission tomography (PET) to noninvasively assess PD-L1 expression in tumors, particularly in the context of breast cancer. The probe consists of the B11 nanobody, a small (~15 kDa) antibody fragment that specifically targets programmed death-ligand 1 (PD-L1), conjugated with a p-SCN-Bn-DTPA chelator and radiolabeled with the positron-emitting isotope scandium-44 (44Sc). While nanobodies typically offer favorable pharmacokinetics for imaging due to their rapid blood clearance and high tissue penetration, in vitro studies of this specific construct revealed high non-specific uptake, which limited its potential as a molecular imaging agent. As a result, the probe was not advanced to in vivo evaluation or clinical development.
